Tirzepatide (LY-3298176): a scientific overview
Tirzepatide is a synthetic once-weekly dual agonist of the GLP-1 and GIP incretin receptors, developed by Eli Lilly and marketed as Mounjaro (type 2 diabetes) and Zepbound (chronic weight management).
For laboratory research use only. Not for human consumption, diagnostic or therapeutic use.
What is Tirzepatide?
Tirzepatide is a 39-amino-acid synthetic peptide with a C20 fatty diacid moiety that confers albumin binding and a plasma half-life compatible with once-weekly subcutaneous dosing. Structurally derived from the native GIP sequence, it activates both GIP and GLP-1 receptors with distinct signalling profiles.
Mechanism of action
- GLP-1 receptor agonism — increases glucose-dependent insulin secretion, delays gastric emptying, and reduces food intake through central appetite pathways.
- GIP receptor agonism — potentiates insulinotropic effects and modulates adipose tissue lipid handling, contributing to improved insulin sensitivity.
- Biased signalling — Tirzepatide shows reduced β-arrestin recruitment at the GLP-1 receptor, associated with better tolerability in dose-escalation studies.
Published trial data
In the SURPASS Phase 3 program (type 2 diabetes), Tirzepatide 5, 10 and 15 mg produced dose-dependent HbA1c reductions of up to about 2.4 percentage points versus placebo and outperformed semaglutide 1 mg (SURPASS-2). The SURMOUNT program (obesity) reported mean body-weight reductions of approximately 15–22% at 72 weeks with 5–15 mg weekly dosing versus placebo (Jastreboff et al., NEJM 2022).
Reconstitution & storage
Lyophilised Tirzepatide is typically reconstituted with bacteriostatic water for injection. Common working concentrations range from 5 to 10 mg per mL, adjusted to vial strength and protocol. Reconstituted peptide should be stored at 2–8 °C, protected from light, and used within 28 days. Unreconstituted vials remain stable long-term at −20 °C.
Quality & purity
Every batch supplied by XCE Peptide is characterised by reversed-phase HPLC and mass spectrometry. Only batches above 99% purity are released, with randomised samples cross-verified by an independent European laboratory. A batch-specific certificate of analysis accompanies every order.
Frequently asked questions
How does Tirzepatide differ from semaglutide?
Semaglutide is a single GLP-1 agonist. Tirzepatide adds GIP receptor agonism, which contributes to greater HbA1c and body-weight reductions in head-to-head studies.
Is research-grade Tirzepatide the same as Mounjaro?
The active molecule is identical, but research-grade lyophilised peptide is supplied without the sterile injectable formulation, preservatives or delivery device of the licensed medicine.
What purity should research-grade Tirzepatide reach?
Research protocols typically require ≥ 98% HPLC purity. XCE Peptide releases at ≥ 99% and provides a certificate of analysis with every order.
